Created on January 25, 1834, Hillsborough County was carved from Monroe and Alachua Counties during the U.S. territorial period (1822-1845). At inception, the county was much larger than its current size. The modern-day counties of Charlotte, DeSoto, Hardee, Manatee, Pasco, Pinellas, Polk, and Sarasota were all part of Hillsborough County.

The county played a significant role during the Seminole Wars as the U.S. government set up Fort Brooke at the edge of the Hillsborough River. The arrival and establishment of phosphate and cigar manufacturing during the 19th century led to a massive rise in the county's population.
